DNA is the most critical genetic material within organisms, and its integrity is directly related to the accuracy of genetic information. However, DNA damage is a common phenomenon within cells, mainly caused by endogenous factors (such as replication errors, oxidative deamination, and reactive oxygen species) and exogenous factors (such as ultraviolet light and radiation), leading to abnormalities in the chemical structure or coding characteristics of DNA. If this damage is not repaired in a timely manner, it may lead to cellular physiological abnormalities. Types of DNA damage include singlestrand breaks (SSBs), double-strand breaks (DSBs), phosphodiester bond breaks, and base damage (such as pyrimidine dimers, mismatches, and crosslinks).
To cope with DNA damage, cells have developed multiple mechanisms, collectively known as DNA Damage Response (DDR), which can detect and promote the repair of DNA damage.
There are several main pathways in DDR:
1.Non-homologous End-joining (NHEJ) 2.Homologous Recombination Repair (HRR) 3.Nucleotide Excision Repair (NER)
4.Fanconi Anemia (FA) 5.Translesion Synthesis (TLS) 6.Base Excision Repair (BER) 7.Mismatch Repair (MMR)
