Pyroptosis is a special form of inflammatory cell death that is crucial for the innate immune defense of the organism, especially in resisting foreign pathogens and responding to
endogenous signals. This form of cell death is closely related to the inflammasome.Characteristics of pyroptosis include nuclear condensation, chromatin fragmentation, and
cellular swelling with bleb formation, ultimately leading to cell membrane rupture and the release of cellular contents, triggering an inflammatory response. Pyroptosis mainly occurs
through two pathways: the classic pathway dependent on Caspase-1 and the non-canonical pathway dependent on Caspase-4/5/11.
To determine whether the induced cell death is pyroptosis, the following indicators need to be detected:
1.The cleavage of GSDMD;
2.The activation level of Caspases;
3.The cleavage and maturation of IL-1β and IL-18 precursors;
4.The detection of chromatin integrity.
