Description PRINCIPLE OF THE ASSAY This assay employs the quantitative competitive enzyme immunoassay technique. Recombinant Human MAPT has been pre-coated onto a microplate. Standards or samples are premixed with biotin-labeled antibody and then pipetted into the wells. Tilavonemab in the sample competitively binds to the pre-coated protein with biotin-labeled Tilavonemab. After washing away any unbound substances, Streptavidin-HRP is added to the wells. Following a wash to remove any unbound enzyme reagent, a substrate solution is added to the wells and color develops in inversely proportion to the amount of Tilavonemab bound in the initial step. The color development is stopped and the intensity of the color is measured.

Stability and Storage When the kit was stored at the recommended temperature for 6 months, the signal intensity decreased by less than 20%.

Background Tilavonemab (ABBV-8E12) is an antibody recognizing the aggregated, extracellular form of pathological tau and binding to the N-terminus of tau. This drug was developed by C2N Diagnostics and AbbVie and has been validated for its safety in a phase I trial. However, the phase II trial, evaluating the efficacy and safety of tilavonemab in 453 patients with early AD, did not obtain expected results and now tilavonemab is discontinued in AD treatment. In addition, a phase II trial purposed to assess the long-term safety and tolerability of tilavonemab in 364 participants with early AD was finished in September 30, 2021, but its final reports are not available.
