Human IL-15 ELISA Kit
Application: For quantitative detection of IL-15 in human serum, plasma, body fluids, tissue lysates or cell culture supernatants.
Introduction: Interleukin 15 (IL-15) is a cytokine that affects T-cell activation and proliferation similarly to IL2. It secreted by mononuclear phagocytes (and some other cells) following infection by virus(es) and normally presented in vivo as a cell-associated cytokine bound to IL15RA. IL-15 binds to and signals through a complex composed of IL-2/IL-15 receptor beta chain (CD122) and the common gamma chain (gamma-C, CD132). In rodent lymphocytes, IL-15 prevents apoptosis by inducing an apoptosis inhibitor, BCL2L1/BCL-x(L). In human s with celiac disease IL-15 similarly suppresses apoptosis in T-lymphocytes by inducing Bcl-2 and/or Bcl-xL. There have been recent studies suggesting that suppression of IL-15 may be a potential treatment for celiac disease and even presents the possibility of preventing its development.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-15 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-IL-15 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-15 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-15 can be calculated.
Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
Range: 46.9 pg/ml - 3000 pg/ml
Sensitivity: < 3 pg/ml
