Human TRAIL ELISA Kit
Application: For quantitative detection of TRAIL in human serum, plasma, body fluids, tissue lysates or cell culture supernatants.
Introduction: TNF-related apoptosis-inducing ligand (TRAIL), is a protein functioning as a ligand that induces the process of cell death called apoptosis. It is encoded by the TNFSF10 gene, which maps to 3q26, spans approximately 20 kb and contains 5 exons. TRAIL shows homology to other members of the tumor necrosis factor superfamily. It is composed of 281 amino acids and has characteristics of a type II transmembrane protein. TRAIL might induce apoptosis of brain tissue, indicating a potential target for treatment of multiple sclerosis. In cells expressing DcR2, TRAIL binding therefore activates NFkappaB, leading to transcription of genes known to antagonize the death signaling pathway and/or to promote inflammation.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-TRAIL polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-TRAIL polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the TRAIL amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of TRAIL can be calculated.
Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
