Human CHI3L1 / YKL-40 ELISA Kit

Human CHI3L1 / YKL-40 ELISA Kit

Size
Application: For quantitative detection of CHI3L1 / YKL-40 in human serum, plasma, body fluids, tissue lysates or cell culture supernatants.

Introduction: Chitinase-3-like protein 1 (CHI3L1), also known as YKL-40, is a secreted glycoprotein that is approximately 40kDa in size that in human s is encoded by the CHI3L1 gene. The name YKL-40 is derived from the three N-terminal amino acids present on the secreted form and its molecular mass. YKL-40 is expressed and secreted by various cell-types including macrophages, chondrocytes, fibroblast-like synovial cells, vascular smooth muscle cells, and hepatic stellate cells. The biological function of YKL-40 is unclear. It is not known to have a specific receptor. Its pattern of expression is associated with pathogenic processes related to inflammation, extracellular tissue remodeling, fibrosis and solid carcinomas[8] and asthma.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-CHI3L1 / YKL-40 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-CHI3L1 / YKL-40 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CHI3L1 / YKL-40 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of CHI3L1 / YKL-40 can be calculated.
