Application: For quantitative detection of BMP-6 in human serum, plasma, body fluids, tissue lysatesor cell culture supernatants.
Introduction: Bone morphogenetic protein 6 is a protein that in humans is encoded by the BMP6 gene.The protein encoded by this gene is a member of the TGFβ superfamily. Bone morphogenetic proteins are known for their ability to induce the growth of bone and cartilage. BMP6 is able to induce all osteogenic markers in mesenchymal stem cells.The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-BMP-6 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-BMP-6 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to theBMP-6amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofBMP-6can be calculated.
Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
