Human IL-1RA / IL-1F3 ELISA Kit

Human IL-1RA / IL-1F3 ELISA Kit

Size
Application: For quantitative detection of IL-1RA / IL-1F3 in human serum, plasma, body fluids, tissue lysates or cell culture supernatants.

Introduction: The interleukin-1 receptor antagonist (IL-1RA) is a protein that in humans is encoded by the IL1RN gene. IL-1RA is a member of the interleukin 1 cytokine family. IL-1RA is secreted by various types of cells including immune cells, epithelial cells, and adipocytes, and is a natural inhibitor of the pro-inflammatory effect of IL1β. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- IL-1RA / IL-1F3 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- IL-1RA / IL-1F3 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-1RA / IL-1F3 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-1RA / IL-1F3 can be calculated.
