Growth factors, steroid hormones, G protein-coupled receptor ligands, and neurotransmitters can initiate MAPK signaling pathways. Activation of ERK1 and ERK2 requires phosphorylation by upstream kinases such as MAP kinase kinase (MEK), MEK kinase and Raf-1.
